Expression of Inflammatory Factors in Fibroblastic Reticular Cell by Tumor Necrosis Factor-¥á

Abstract
Fibroblastic reticular cells (FRC) play tissue reorganization roles in lymph node (LN). The novel concept of FRCs as inflammatory modulators was documented by present study. By establishing an in vitro experimental model containing FRC, we reported that FRC produce inflammatory materials, which regulate biological functions of T cells. To investigate the production of cyclooxygenase (COX)-1, 2 and inducible nitric oxide synthase (iNOS) in FRC, we performed Western blot technology. In order to examine inflammatory cytokines, DNA microarray was performed. Moreover, to observe translocation of nuclear factor kappa-B (NF-¥êB) into nucleus, immunofluorescence technology was applied. COXs and iNOS productions were detected and up-regulated by tumor necrosis factor-¥á (TNF-¥á) in FRC. Several inflammatory cytokines were induced by TNF-¥á. NF-¥êB in FRC stimulated by TNF-¥á was translocated into nucleus. The current results suggest that FRC may help understand the potential effects of inflammation and immune response.
